I take creatine, and most of the time I do not notice it. The day it gets noticed is the morning after a short night: I feel less tired, roughly. Not awake the way coffee makes you, which I do not drink because it makes me fast and bad. Less damaged.
I had filed that under impressions, until two recent trials measured exactly that situation: a night without sleep, a dose of creatine, tests every two and a half hours, and a brain spectroscopy to see what happens inside. They find an effect. They also find a dose that has nothing to do with the 5 g on the box.

Why the brain runs short of energy when it does not sleep
Creatine serves to recharge the cells' energy molecule, ATP, when demand exceeds what cellular respiration supplies on the spot. In muscle, that is a sprint. In the brain, it is all the time, and more so when you force it to stay awake.
That is the starting point of the 2024 trial. Its authors recall that sleep deprivation lowers the brain's high-energy phosphates and the creatine it contains, and that supplementation does the opposite. The problem is that the brain absorbs very little creatine from outside: until now, weeks of daily intake were needed to measure a rise.
Their hypothesis fits in one sentence: if you flood the blood with creatine at the precise moment the brain is burning its reserves, the passage opens temporarily. Hence a single, huge dose, taken at the start of a sleepless night.
What the 2024 trial measured
Fifteen healthy adults, eight women, 23 years old on average, each going through creatine and placebo, double-blind. The dose: 0.35 g per kilo of creatine monohydrate, 24 g for a 70 kg person. Then 21 hours awake, with cognitive tests and two brain spectroscopies at baseline, then 3, 5.5 and 7.5 hours after intake.
On creatine, the brain energy markers move the right way: the phosphocreatine to phosphate ratio goes back up, ATP too, brain creatine increases, and pH does not drop as it does under placebo. Cognitive performance and processing speed are better than under placebo. The maximum effect comes four hours after intake, and creatine was well tolerated.
The authors conclude that a single high dose can partially reverse the metabolic alterations and the fatigue-related cognitive deterioration. Partially. Nobody becomes rested again.
And with a more reasonable dose?
That is the question the same team asked in 2026, with 29 participants and a reduced dose of 0.2 g per kilo, 14 g for 70 kg, in the same 21-hour protocol. The effect is still there: less deterioration on logic and numerical tasks, on language processing speed and on the psychomotor vigilance test, with up to 12% improvement. Less pronounced than with the high dose, but present. Women benefited more than men on several tasks.
Two doses, two trials, one direction. And one shared limit: neither tested 5 g. The dose that produces the sleepless-night effect is three to five times the one you take for muscle.
What the earlier trials said
Before these two trials, the literature rested on another logic: load for a week, then deprive of sleep. McMorris, in 2006, gave 5 g four times a day for seven days, 20 g per day, to ten people against nine on placebo, then 24 hours awake with light exercise. At 24 hours, the creatine group had deteriorated less on random movement generation, reaction time, balance and mood.
The following year, same protocol, 36 hours awake this time: creatine did better on only one task, the most demanding for executive function, and only at 36 hours. Nothing on short-term memory, reaction time or mood. Twenty grams a day for a week therefore protects little, and only what costs the brain the most.
The third trial is the closest to real life. Cook, in 2011, took ten professional rugby players, had them sleep between three and five hours, and gave them 1.5 hours before a passing test either a placebo, or 50 or 100 mg per kilo of creatine, 3.5 to 7 g for 70 kg, or 1 or 5 mg per kilo of caffeine. On placebo, accuracy collapses. On creatine as on caffeine, at any of the doses, it does not move. Useful detail: high-dose caffeine raised salivary cortisol, creatine did not.
Outside sleep deprivation, the effect is small
This matters to avoid over-reading. Prokopidis's meta-analysis, in 2023, eight trials, finds a small effect of creatine on memory, clear in 66 to 76 year olds and nil in 11 to 31 year olds. A letter then flagged a statistical double count likely to have produced a false positive. Xu's meta-analysis, in 2024, over 16 trials and 492 participants, finds a small effect on memory with certainty rated moderate, an effect on attention and processing speed with low certainty, and nothing on overall cognition or executive function.
A 2026 review sums up the field with a title that says it all: have we put the cart before the horse? Measuring brain creatine depends heavily on method, the rise under supplementation depends on dose and duration, and the most solid benefits are seen during acute metabolic stress: sleep deprivation, mental fatigue, lack of oxygen. The rest is preliminary.
In a rested young adult, there is no deficit to fill. That is precisely why I notice nothing most of the time, and something after a short night.
What my observation is worth
There is a difference I cannot settle alone. The trials measure a single high dose taken before a sleepless night. I take a maintenance dose every day, and I notice something after a short night. Those are two possible mechanisms: brain reserves a little fuller thanks to chronic intake, or an impression. The meta-analyses say the first exists, but that it is small at my age.

In practice: the maintenance dose every day, because that is where the literature is widest. A single high dose only for a sleepless night I cannot avoid, knowing that 24 g at once has only been tested in 15 people. And never as an excuse to shorten the next night.
What this article does not say
- That 5 g in the morning after a short night does anything. No trial has tested it. The single doses that work are three to five times higher.
- That creatine replaces sleep. It absorbs part of the drop during forced wakefulness, in the laboratory. Nobody measured the next day, nor repeated short nights.
- That my impression is an observation. No alternation, no measurement, and a dose that is not the one in the trials.
- That 24 g at once is harmless. The trial notes good tolerance in 15 young adults. That is not safety data, and creatine inflates the creatinine of a blood test.
- That the effect on memory is settled. Small, mostly after 65, and the most cited meta-analysis has been challenged for a counting error.
